Miyamoto Musashi is the protagonist of Onimusha: Way of the Sword. Modeled after Toshiro Mifune, voiced by Yoshimasa Hosoya (JP) and Kenichiro Thomson (EN), he wields the Oni Gauntlet in a corrupted Edo-era Kyoto.

Izumo No Okuni is a confirmed ally in Onimusha: Way of the Sword. The historical founder of kabuki, reimagined as a young dancer who has left the stage to hunt the Genma destroying Kyoto.

Dokyo is a sinister antagonist in Onimusha: Way of the Sword. She operates a secret underground laboratory beneath Edo-era Kyoto where she creates new Genma variants and drives the spread of Malice across the city.

Lady Oni is a female Oni housed inside Musashi's Oni Gauntlet in Onimusha: Way of the Sword. She speaks to him from within the gauntlet, guides him through corrupted Kyoto, refuses to disclose her full identity, and can manifest outside the gauntlet when needed.

Ono No Takamura is a confirmed ally in Onimusha: Way of the Sword. Reimagined from the Heian-era scholar and poet, he is an old master swordsman and the third confirmed Oni Gauntlet bearer alongside Musashi and Ganryu.
