Insomniac has presented a closed set of antagonists for Marvel's Wolverine: Bolivar Trask, Omega Red, Deathstrike, the Reavers, and The Hand. Because the story is original, that list is the boundary. A villain who exists in the comics is not in this game until the game shows them.
Trask is an anti-mutant billionaire industrialist driven by a fanatical belief in human superiority, and he drives the campaign. He pays the Reavers to abduct mutants, deploys Ability Inhibitors against Team X, builds the Sentinel Prototype, and constructs a fake town for one of his experiments. Official material describes him as an industrialist rather than a scientist, and no doctorate or research role has been confirmed for him.
The Reavers are a guns-for-hire cybernetic militia working for Trask, and they are the enemy Logan meets most often. They carry out the mutant abductions the story is built around, and previews describe their weapons as electrically charged blades.

Attacks that cannot be blocked are telegraphed with a red tinge. The accessibility settings include colour shaders for seven different enemy types, which is the clearest published indication of how many distinct enemy classes ship.
The Hand is a new enemy faction for this game, wearing the trademark red garb. Insomniac has said Logan and The Hand share a history that players uncover as they tear through the story, and has deliberately not revealed what it is.
In combat they fight very differently from the Reavers: fluid acrobatics, precise projectiles, and special blades that pierce Wolverine and temporarily stunt his healing. Individual members carry katanas or bows, and a Critical Strike can take that weapon and slam it back into them.
They also drive one of the campaign's set pieces. The Hand kidnap Walter Langowski, which sets off a rooftop chase across Madripoor.
Ability Inhibitors are Trask technology rather than an enemy type. They block Wolverine and his teammates from using their abilities, which strips Logan of his healing factor for as long as they are active. Insomniac's framing is that he has to work past the pain and fight without healing, destroying the Inhibitors before it is too late. Previews have also called them mutant-gene inhibitors and described them as creating flashes of vulnerability in otherwise one-sided fights.
The Sentinel Prototype is a boss built by Trask to detect and destroy mutants. It is an imposingly large mechanical creation mounted to a wall and has no legs, which does not stop it from covering the arena: it ground pounds, fires scorching lasers and missile barrages, punches, and sweeps with forearm slaps.
The fight is designed around the whole squad. Insomniac describes it as needing the combined abilities of Team X, with Mystique available as a commandable ally, and it ends with Logan bursting through the machine's head. In-game dialogue presents it as an expanded version of a Sentinel encountered during an earlier mission set in Vancouver.
Omega Red is confirmed as a villain and is described in official copy as boorish. Logan has a run-in with him during the opening Telambang mission, on the way to freeing Nathaniel Essex. Nothing beyond that appearance has been detailed.
Deathstrike has been described by Insomniac as one of the game's big antagonists and appears in a cinematic trailer. That is the extent of what has been confirmed: no role, no fight, and no abilities have been published, and the character is uncast in public material.
The name appears in two forms. Insomniac uses Deathstrike, while some licensor material uses Lady Deathstrike. This wiki follows the developer's wording and treats the longer form as a variant.
Stealth does not work against bosses. Boss encounters are open fights from the start, and a separate accessibility toggle controls whether the boss health bar is displayed. See Combat and Accessibility Options.
